Pippa Middleton and her husband must dig up a car park on their Kintbury estate after a government planning inspector backed West Berkshire Council's refusal.
The couple had sought retrospective permission for the site on their Barton Court Estate after the car park was built last year, but an appeal was also refused.
"Due to its size and engineered appearance the development has introduced a visually intrusive and urbanised feature in to an otherwise open meadow," said the planning inspector Alexander O'Doherty.
The car park sits on an old paddock, north of the River Kennet, in the Wessex Downs - an area with enhanced protections as a National Landscape and particular scenic beauty.
Agents said Barton Court has a long history of fly fishing and that the leisure use provides income for upkeep, but they did not supply financial evidence. Concerns were also raised that vehicles leaving the car park would not have adequate visibility.
Local Neil Warner told planners there were plenty of footpaths used by anglers and said he was 'shocked' when the 'ugly' fence was put up.
At a public inquiry earlier this year Mr Matthews argued privacy fears and the family's high profile required tighter security. That inquiry is still to report its findings and was due in August (2026).
"The ground should be reinstated forthwith to its former marsh status and planted with appropriate plants.
"Barton Court is a large estate, surely they have hardstanding on their own premises for anglers."
